Characterisation of the Molecular Basis of Systemic Salmonellosis in Calves

Objective

<ol> <LI> To further characterise the mechanisms of systemic translocation of Salmonella at time points following oral inoculation of calves; <LI> Togenerate a signature-tagged transposon bank for Salmonella enterica serovar Dublin and select mutants proficient in systemic spread; <LI> To select tagged mutants containing transposon insertions in selected genes; <LI> To screen selected tagged mutants of serovars Dublin and Typhimurium within bovine tissues and efferent lymphatics to determine whether distinct virulence loci contribute to systemic translocation in calves; <LI> To construct defined mutations in genes important for systemic salmonellosis, and confirm their role in calves by trans-complementation studies.
