Definition of targets to inhibit Salmonella colonization of the intestine

Objective

As the budget from this proposal was reduced from $2.88 million to $1.65 million (a 43% cut), the objectives of this proposal are revised accordingly (see below).Specific Aims:Aim 1. Quantify the survival and growth of candidate mutants in anaerobic growth conditions in vitro and in calves.This aim will undergo only a minor change: gene expression studies during oxygen limitation will not be performed (B.4.a.iii.a)Aim 2. Determine which genes influence known virulence pathways.Ligated ileal loop experiments in this aim will not be performed (B.4.b.iii.b)Effector secretion studies (B.4.b.iii.c) will be limited to those mutants with demonstrable effects on TTSS-1, and TTSS-2 machinery or effector gene expression as identified in B.4.b.iii.a.Aim 3. Define new paradigms in pathogenesis by identifying functional pathwaysWe will limit transcriptional analysis and follow up experiments to one putative regulator in this aim (instead of 2).We will test point mutants in selected conserved domains for function in vitro, but will test these mutants in calves only as funds permit.Aim 4. Model structure and identify specific substrates for selected candidate genesWe will limit modeling of structure and substrate identification to one protein in this aim (instead of 2). Additional modeling will be performed if funds are available.
