DEVELOPMENT OF A GENETIC PERTURBATION PLATFORM FOR GRAM-POSITIVE BACTERIA

Objective

Objective 1. Optimization of protein electroporation conditions with Leuconostoc citreum, a representative Gram-positive bacteria. In order to easily evaluate electroporation conditions, we will use fluorescently-labeled BSA as a target protein (FITC-BSA). Electroporation efficiency will be evaluated using flow cytometry and fluorescence microscopy. Conditions will be tested with a full factorial design and will include the number of cells, voltage, and resistance.
