Insect Management for Reduction of Mycotoxigenic Fungi in Midwest Corn

Objective

<OL> <LI> Discover, characterize, and introduce novel insect resistance mechanisms, alone and in combination, to corn through molecular biological approaches. Mechanisms emphasized will be those potentially active against plant pathogens whenever possible and directed primarily towards ear-damaging Midwestern insects. <LI>Develop comprehensive management systems for mycotoxins in Midwest corn, which will include predictive models, insect monitoring and control, resistant varieties, and cultural practices, etc.

Use bioassay guided fractionization, empirical testing of available proteins and allelochemicals, and protein expression systems to isolate bioactive materials in determining new resistance mechanisms. Proteins and nucleic acids will be purified using conventional techniques such as gel filtration, diferential precipitation, size exclusion and affinity chromatography, electrophoresis, hybridization, PCR, blotting, commercial kits, etc. Use in vitro bioassays, transient, and stable expression to determine effective combinations of resistance mechanisms. Use integrated management scheme including sanitation, rotation, resistant hybrids, insect monitoring for insecticide application decision, predictive models, ear sampling, and mycotoxin analysis (HPLC by commercial laboratories), biological control, insect dispersal of antifungal organisms, yield determination, and cost benefit analysis in representative commercial fields.
